Gene Silencing RNA Nanoparticles

Next-generation targeted delivery vehicles combining multivalent siRNA payloads with active targeting aptamers on self-assembling RNA nanostructures.

Targeted Nanomedicine

Overcoming the delivery barrier for therapeutic RNAi

While siRNA therapeutics have transformed targeted medicine, conventional delivery platforms often suffer from non-specific liver accumulation, poor biodistribution to extrahepatic tissues, or lipid nanoparticle (LNP)-induced immunogenicity.

RNA Nanorobotics engineers programmable, scaffold-based RNA nanoparticles that carry both therapeutic siRNA payloads and cell-surface receptor targeting ligands on a single, biocompatible macromolecule. Our platform enables cell-specific gene knockdown in solid tumors, immune cells, and difficult-to-transfect targets.

Nanoparticle Modular Composition

  • 01
Core Thermodynamic Scaffold: Ultra-stable 3WJ (three-way junction), tetrahedral, or planar RNA motifs engineered to remain intact in physiological conditions without dissociation.
  • 02
Multivalent Gene Silencing Payloads: Multiple siRNA or DsiRNA branches targeting single or synergistic oncogenes, viral genes, or disease drivers.
  • 03
Targeting Ligands & Imaging Moieties: RNA aptamers (e.g., EGFR, PSMA, transferrin), small molecules (folate, GalNAc), or fluorophores (Cy5.5, Alexa Fluor) for non-invasive biodistribution imaging.

Platform Features

Engineering & Characterization Rigor

Multivalent Therapeutic Architecture

Self-assembling nanoscale scaffolds displaying multiple siRNA duplexes alongside targeting aptamers or chemical ligands (folate, GalNAc, peptides) with precise stoichiometric ratios.

Ultra-High In Vivo Stability

Constructed using 2′-fluoro, 2′-O-methyl, and 3WJ core scaffolds that resist serum RNase degradation for >24 hours and maintain thermodynamic stability in blood circulation.

Receptor-Mediated Cell Entry

Aptamer- and ligand-directed targeting promotes active receptor-mediated endocytosis, facilitating intracellular release of active siRNA payloads directly into the cytoplasm.

Favorable Pharmacokinetics & Low Immunogenicity

Nanoscale dimensions (10–50 nm) enable enhanced EPR accumulation at disease sites, escape rapid renal clearance, avoid hepatic entrapment, and exhibit zero interferon stimulation.

Comprehensive Analytical Validation Package

Every batch of Gene Silencing RNA Nanoparticles ships with a full quality control package generated using our in-house instrumentation:

Native PAGE Gel Confirmation of 100% self-assembly efficiency and absence of free uncomplexed monomer strands.
DLS & Zeta Potential Precise hydrodynamic size distribution (monodisperse polydispersity index <0.2) and surface charge.
Serum Nuclease Assay Incubation in 50% fetal bovine serum or human serum demonstrating biological stability over 24–48 hours.
HPLC / ESI-MS Chemical purity validation and sequence confirmation of all synthetic building blocks.
